چکیده

Polyunsaturated lipids are an essential component of biological membranes, influencing order and dynamics of lipids, protein-lipid interaction, and membrane transport properties. To gain an atomic level picture of the impact of polyunsaturation on membrane properties, quantum mechanical (QM) and empirical force field based calculations have been undertaken. The QM calculations of the torsional energy surface for rotation about vinyl-methylene bonds reveal low barriers to rotation, indicating an intrinsic propensity toward flexibility. Based on QM and experimental data, empirical force field parameters were developed for polyunsaturated lipids and applied in a 16 ns molecular dynamics (MD) simulation of a 1-stearoyl-2-docosahexaenoyl-sn-glyerco-3-phosphocholine (SDPC) lipid bilayer. The simulation results are in good agreement with experimental data, suggesting an unusually high degree of conformational flexibility of polyunsaturated hydrocarbon chains in membranes. The detailed analysis of chain conformation and dynamics by simulations is aiding the interpretation of experimental data and is useful for understanding the unique role of polyunsaturated lipids in biological membranes. The complete force field is included as Supporting Information and is available from http://www.pharmacy.umaryland.edu/faculty/amackere/research.html.

The physicochemical and organoleptic evaluation of the nano/micro encapsulation of Omega-3 fatty acids in lipid vesicular systems

Objective(s): Omega-3 fatty acids play a key role in maintaining human health. The present study aimed to reduce the fishy smell and taste of omega-3 fatty acids through the encapsulation of lipid vesicles. Materials and Methods: Different non-ionic surfactants from the sorbitan ester family and egg lecithin with cholesterol were utilized to form micro-niosomal and liposomal formulations ...

Lipid bilayer regulation of membrane protein function: gramicidin channels as molecular force probes.

Membrane protein function is regulated by the host lipid bilayer composition. This regulation may depend on specific chemical interactions between proteins and individual molecules in the bilayer, as well as on non-specific interactions between proteins and the bilayer behaving as a physical entity with collective physical properties (e.g. thickness, intrinsic monolayer curvature or elastic mod...

Docosahexaenoic acid alters bilayer elastic properties.

At low micromolar concentrations, polyunsaturated fatty acids (PUFAs) alter the function of many membrane proteins. PUFAs exert their effects on unrelated proteins at similar concentrations, suggesting a common mode of action. Because lipid bilayers serve as the common "solvent" for membrane proteins, the common mechanism could be that PUFAs adsorb to the bilayer/solution interface to promote a...

Metabolic Effects of Polyunsaturated Fatty Acids in Chickens: A Review

Chicken has been used as a suitable model for lipid metabolism studies, because dietary modifications especially dietary fat type can change chicken body composition. Fats act as a condense source of energy and certain fatty acids such as polyunsaturated fatty acids (PUFAs) are required for both animal and human health. The n-3 PUFAs, especially, eicosapentaenoic acid (EPA) and docosahexaenoic ...

Omega-3 Long Chain Polyunsaturated Fatty Acids as Sensitizing

The efficacy of chemotherapy depends on sensitivity and intrinsic or acquired drug 14 resistance of cancer cells. The n-3 long chain polyunsaturated fatty acids (n-3 LCPUFAs) are 15 considered chemosensitizing agents and revertants of multidrug resistance by pleiotropic 16 mechanisms. The specific mechanisms are not fully understood, but nowadays, it is widely accepted 17 that there are a compl...
